My wife and I are planning to camp in the Yosemite area in mid- September 2011. We'll be towing a 33 ft fifth wheel and was wondering which is the best route to take from I-80 to Yosemite. Also would like recommendations on good campgrounds in the area with full hook-ups. Tell us your favorites.

Come in to Yosemite from the east otherwise you'll have to go across to Sacramento and come down HWY-99.
From the east- I-80 to US 95 south to NV-359 which changes to CA-167 From CA-167 take HWY-395 south and then head west on CA-120 Tioga Pass to get to Yosemite.
If you plan on staying in the park be sure to go on the website and see what the rules are for RVs, some have changed due to cutbacks and to limit park use.
We stayed here, it was nice - http://koa.com/campgrounds/yosemite-south/ But is on the west side of Yosemite.
There's not much to choose from on the east side of the park for RV parks.

If you are a member of Escapees there is the Park of the Sierras. We paid $97 a week plus electric this past March. The most unique rv park we have been to. It would be worth it to join Escapees just to use this park. I think it was about 25 minutes to the entrance of Yosemite.
